はゴルジ体のシス層表面に局在するタンパク質であり,輸送小胞とゴルジ体の繋留と融合,ゴルジ体の層板構造維持に重要な役割を果たしていると考えられている。以前にGM130 欠損CHO 細胞が温度感受性となり39.5̊C ではゴルジ体が分散し,ゴルジ体を経由した小胞輸送経路が停止して致死となることが報告されていた。一方最近,ヒト遺伝病の研究やGM130 遺伝子破壊マウスの解析から,GM130 欠損は細胞レベルでは致死ではないが,神経系の異常や精子形成異常を示すことが報告された。これらのケースでは,GM130 遺伝子のフレームシフトにより,N 末端300 ~ 400 アミノ酸残基の発現が残存している可能性がある。一方,先のGM130 欠損CHO 細胞ではGM130 欠損の原因変異が特定されていないため,GM130 欠損は致死ではなく,GM130 欠損以外の原因で温度感受性致死となっていた可能性が示唆された。そこで本研究ではCRISPR/Cas9 法を用いて培養細胞レベルでGM130 遺伝子破壊を行い,この可能性を検証することを試みた。Ann Ran らによって開発されたプラスミドと薬剤選択を用いるCRISPR/Cas9 法によりHEK293 細胞とHeLa 細胞でGM130 遺伝子破壊を行った。その結果,GM130 陰性細胞の生成を確認することができたが,これらの細胞のほとんどがアポトーシスにより死滅することが明らかとなった。このことから,GM130
