N-acetylgalactosaminyltransferase (GalNAc-T) は,その生合成開始反応を触媒する.筆者らはこれまでに,脳・神経特異的に発現するGalNAc-T9および,それと非常に高い相同性をもつGalNAc-T様遺伝子(putative GalNAc-T,以降pt-GalNAc-Tと略)をクローニングした.pt-GalNAc-Tについては,ノーザンブロット解析,in situ hybridization 解析により,それがGalNAc-T9同様,脳特異的に発現することを見いだしている.pt-GalNAc-Tが典型的なムチン型ペプチドに対して糖転移活性を示すことから,pt-GalNAc-TはGalNAc-Tファミリーの新規アイソザイムとみなすことができるので,GalNAc-T16と呼ぶことにする.筆者らは,GalNAc-T9および-T16の生体内での機能を詳細に解析するため,胚性腫瘍細胞P19を用いた.P19はレチノイン酸(RA)存在下で神経に分化する細胞であり,神経分化モデル細胞として様々な研究に用いられている.まず,P19細胞におけるGalNAc-Tファミリーの発現を解析したところ,神経特異的に発現するGalNAc-T9,-T13,-T16の発現が神経分化に伴って亢進することを見いだした.そこで,P19細胞においてこれらの遺伝子の発現を抑制し,神経分化に対する影響を解析した.興味深いことに,GalNAc-T16の発現を抑制したP19細胞においてのみ,神経細胞への分化が見られず細胞死を引き起こした.筆者らはさらに,カスパーゼ解析,FACS解析により,GalNAc-T16ノックダウン細胞における神経分化誘導時にアポトーシスを起こしていることを見いだした.以上の結果から,GalNAc-T16は神経分化において重要な機能を持つと考えられた.","subitem_description_type":"Abstract"}]},"item_10002_description_6":{"attribute_name":"内容記述","attribute_value_mlt":[{"subitem_description":"1.序論\n2.材料と方法\n3.結果\n4.考察\n参考文献","subitem_description_type":"Other"}]},"item_10002_publisher_8":{"attribute_name":"出版者","attribute_value_mlt":[{"subitem_publisher":"京都産業大学"}]},"item_10002_source_id_11":{"attribute_name":"書誌レコードID","attribute_value_mlt":[{"subitem_source_identifier":"AA11923897","subitem_source_identifier_type":"NCID"}]},"item_10002_source_id_9":{"attribute_name":"ISSN","attribute_value_mlt":[{"subitem_source_identifier":"1348-3323","subitem_source_identifier_type":"PISSN"}]},"item_10002_version_type_20":{"attribute_name":"著者版フラグ","attribute_value_mlt":[{"subitem_version_resource":"http://purl.org/coar/version/c_970fb48d4fbd8a85","subitem_version_type":"VoR"}]},"item_creator":{"attribute_name":"著者","attribute_type":"creator","attribute_value_mlt":[{"creatorNames":[{"creatorName":"鳥羽,
