Plastids are thought to have unique DNA replication systems, where DNA primase and DNA helicases are responsible for synthesis of primers and DNA unwinding, respectively. While earlier studies have identified DNA primase and helicase activities in a plastid fraction but little is known about genes encoding the plastidial DNA pirmase. Here we characterized a rice homologue of TWINKLE(Oryza sativa cv. Nipponbare, termed OsTWINKLE), which is an autosomal dominant progressive external ophthalmoplegia(adPEO)-associated gene that encodes mitochondrial DNA helicase in mammalian cells.
