Plastids are thought to have unique DNA replication systems, where DNA primase and DNA helicases are responsible for synthesis of primers and DNA unwinding, respectively. While earlier studies have identified DNA primase and helicase activities in a plastid fraction but little is known about genes encoding the plastidial DNA pirmase and DNA helicase.\n In animal cells, TWINKLE protein, which is is homologue of bacteriophage T7gp4, is known to be localized to mitochondria. While T7gp4 has both DNA primase and helicase activities, animal TWINLE protein has only DNA helicase activity due to sequence change in DNA primase domain. Plant genome encodes TWINKLE protein, and our preliminary subcellular localization analysis showed that rice TWINLE protein is targeted to plastid. Amino acid sequence comparison and homology modeling analysis showed that the plant TWINKLE proteins retain sequence homology for DNA primase domain of the T7gp4.
